Description of Issue

I can output sound from Roon to the internal speakers on the MBP. When I plug headphones into the headphone jack, or run the headphones through a headphone amp connected to the headphone jack, the output still comes out of the speakers, even though the headphone output is selected in System Preferences. But if I play a YouTube video, the sound is routed to the headphones.

When you plug your headphones in, take a look at the settings → Audio page. Do the headphones show up as a separate Audio Device? If so, then you would have to enable that device and then choose it as the output in Roon.
