Probably showing my lack of knowledge and how the DSP engine works, but here goes. My question is. I have a MS600 connected to my amp via analogue. Can I do anything in the Roon’s DSP engine to improve the sound?

Depends what you mean by “improve”.
The MS600 upsamples. You could try different upsampling filters in Roon and send 88/96 to the endpoint which will then simply be converted to analogue. You might prefer a different sound.
You can also fiddle with the parametric eq in Roon if you want a different tonal balance.
Personally I liked the sound of the MS600 as is very much. I felt it to be quite neutral and faithful to the source. YMMV
