I am new to Roon. My setup:
Mac mini M2 playing rune server through this dac: HoloAudio Spring 2 Level 3 Kitsuné edition
2 Airplay speakers
1 bluetooth Speaker
HOW, if at all, can I play through all my speakers at once?

See this FAQ for details about grouping zones. Not all zones can be grouped together, as you will see from the FAQ they need to use the same protocol to be grouped. RAAT zones can be grouped with other RAAT zones and Airplay with other Airplay etc.
I’ve shifted your query into Software Discussion, where you will find a lot of other folks able to help with familiarity and experience . Support is for when things are not working properly, which hopefully, will never happen for you!
So if my main high end stereo is connected by usb, there is no way to play to multiple other speakers simultaneously? (Airplay and Bluetooth) even using a software such as airfoil?
This is a big constraint for me.

It’s not possible in Roon, see the possible zones on the page that was linked above. If you output Roon to a local software device, such as maybe Airfoil might provide, this software can do what it wants, but it’s outside of Roon.
Within Roon, you’d have to find a way to make all devices speak a protocol that can be grouped within Roon.
Bluetooth is something done by your Server and/or Remote computer (hardware feature) and as such transparent to Roon (you play to the device that has Bluetooth active to send the audio to whatever speaker). Instead of using AirPlay directly through Roon, you could do the same with it as with Bluetooth (send the audio to a device that has AirPlay active to send the audio to whatever speaker). If you do that then Roon will not know about resulting delays and may offer only limited possibilities to correct delays if at all. But try it out if you can and if it works for you then it works.
